Enhanced Regeneration Through ex vitro Rooting and Agrobacterium-mediated Genetic Transformation of Eggplant (Solanum melongena L.)

Regeneration of in vitro multiple shoots was achieved through organogenesis on MS supplemented with 2.0 mg/l BAP and 0.5 Kn from cotyledonary leaf explants two local varieties eggplant (Solanum melongena L.). Elongation regenerated obtained growth regulator free MS. In root induction excised less effective or without plant regulators. On the other hand treated 10 mM IBA for 5 min were found to be ex rooting sterilized soil. Following sufficient development roots, rooted plantlets acclimatized room condition, transferred field having 100% survival rate. The regeneration system developed utilized Agrobacterium-mediated genetic transformation using Agrobacterium tumefaciens strain LBA4404/pBI121 containing GUS nptII genes. Adequate response segments bacterial suspension an optical density 0.50 at 600 nm 30 incubation followed by co-cultivation period 72 hrs Nayantara (BARI Begun-5) variety eggplant. Selection transformed carried out BAP, Kn, 300 carbenicillin 100 kanamycin. Stable integration genes confirmed PCR analysis genomic DNA isolated shoots.
